Expanding the Reach of Targeted Therapy

TwoStep Therapeutics leverages tumor-targeting technology based on a unique and modular Polyspecific Integrin-binding Peptide (PIP), which enables delivery of various payloads to virtually any solid tumor.

Multi-Specific Targeting

Unlike traditional agents, PIP binds selectively to five different integrin targets highly expressed on solid tumors. This unique multi-targeting feature allows PIP to target a wide range of tumor types, overcoming the limitations of single-antigen targeting approaches.

Precision and Safety

PIP preferentially binds to integrin conformations abundant in tumor tissue, enhancing specificity and safety.

Integrins are a superfamily of heterodimeric cell-surface adhesion receptors that mediate interactions between cells and the extracellular matrix (ECM). Each integrin is a heterodimer composed of a single α subunit paired with a β subunit, with humans having 18 α and 8 β subunits that combine to form 24 distinct integrins.

Among these, 8 integrins can recognize the Arg-Gly-Asp (RGD) peptide motif found within certain ECM proteins. Many of these RGD-binding integrins are expressed at high levels on tumor cells and their vasculature and facilitate angiogenesis, invasion, and metastasis of numerous cancers. TwoStep’s targeting agent, PIP, is able to bind strongly and selectively to multiple different tumor-associated, RGD-binding integrins, including αvβ1, αvβ3, αvβ5, αvβ6, and α5β1.
